ABSTRACT

Abstract Anabolic substances have been increasingly used by bodybuilders and athletes with the goal of improving performance and aesthetics. However, this practice has caused some concern to physicians and researchers because of unknowledge of consequences that the indiscriminate and illicit use of these substances can cause. Thus, this study analyzed the effects of two commercially available anabolic steroids (AS), Winstrol Depot® (Stanozolol) and Deposteron® (Testosterone Cypionate), in the neuronal density of limbic, motor and sensory regions on the cerebral cortex and in CA1, CA2, CA3 regions of the hippocampus. A total of 60 Swiss mice were used (30 males and 30 females), separated into three groups: control and two experimental groups, which received the AAS. From each brain, homotypic and semi-serial samples were taken in frontal sections from areas established for the study. The results showed that females treated with testosterone cypionate presented a reduction in all regions tested and the ones treated with Stanozolol showed a decrease in some hippocampal areas. Regarding male animals, stanozolol led to a decrease in neuron number in one hippocampal region. These data allow us to conclude that supra-physiological doses of steroids used in this study, can cause considerable damage to nervous tissue with ultrastructural and consequently behavioral impairment. These changes could interfere with the loss of physical yield and performance of athletes and non-athletes and may cause irreparable damage to individuals making irresponsible use of anabolic steroids.

No entanto, essa prática tem causado algumas preocupações aos médicos e pesquisadores, devido ao desconhecimento das consequencias que o uso indiscriminado e ilícito dessas substâncias podem causar. Diante disso, este estudo analisou os efeitos de dois esteroides anabolizantes (EA) comercialmente disponíveis, Winstrol Depot® (Stanozolol) e Deposteron® (cipionato de testosterona), na densidade neuronal das regiões corticais límbica, motora e sensitive bem como das áreas CA1, CA2, CA3 do hipocampo. Foram utilizados 60 camundongos Swiss (30 machos e 30 fêmeas), separados em três grupos: controle e dois grupos experimentais, que receberam o EA. De cada cérebro, foram coletadas amostras homotípicas e semi-seriadas em cortes frontais das áreas estabelecidas para o estudo. Os resultados mostraram que as fêmeas tratadas com cipionato de testosterona apresentaram uma redução em todas as regiões analisadas já as fêmeas tratadas com Stanozolol mostraram uma diminuição em algumas áreas do hipocampo. Em relação aos animais machos, o stanozolol levou a uma diminuição na densidade neuronal em uma região do hipocampo. Estes dados nos permitem concluir que doses supra fisiológicas de esteroides utilizadas neste estudo podem causar danos consideráveis ao tecido nervoso com comprometimento ultraestrutural e consequentemente comportamental. Essas alterações podem interferir na perda de rendimento físico e no desempenho de atletas e não atletas e podem causar danos irreparáveis a indivíduos que fazem uso irresponsável destes EA.

ABSTRACT

Abstract The objective of this study was to verify the effects of ethanol consumption and alcohol detoxification on the biomechanics, area and thickness of cortical and trabecular bone in rat femur. This was an experimental study in which 18 male Wistar rats were used, with 40 days of age, weighing 179±2.5 g. The rats were divided into three groups (n=06): CT (control), AC (chronic alcoholic), DT (detoxification). After experimental procedures, the animals were euthanized by an overdose of the anesthetic and their femurs were collected for mechanical testing and histological processing. All animals did not present malnutrition or dehydration during experimentation period. Morphometric analysis of cortical and trabecular bones in rat femurs demonstrated that AC animals showed inferior dimensions and alcohol detoxification (DT) allowed an enhancement in area and thickness of cortical and trabecular bone. Material and structural properties data of AC group highlighted the harmful effects of ethanol on bone mechanical properties. The results of this study demonstrated that chronic alcoholic rats (AC) presented major bone damage in all analyzed variables. Those findings suggested that alcohol detoxification is highly suggested in pre-operative planning and this corroborates to the success of bone surgery and bone tissue repair. Resumo O objetivo deste estudo foi verificar os efeitos do consumo de etanol e da desintoxicação alcoólica sobre a biomecânica, área e espessura do osso cortical e trabecular em fêmur de ratos. Este foi um estudo experimental no qual foram utilizados 18 ratos Wistar machos, com 40 dias de vida, pesando 179±2,5 g. Os ratos foram divididos em três grupos (n=06): CT (controle), AC (alcoolista crônico), e DT (desintoxicado). Após os procedimentos experimentais os animais foram eutanaziados por uma overdose de anestésico e os fêmures coletados para os testes mecânicos e processamento histológico. Todos os animais não apresentaram desnutrição ou desidratação durante o período de experimentação. As análises morfométrica do osso cortical e trabecular demonstraram que os animais do grupo AC apresentavam dimensões inferiores, enquanto nos animais do grupo DT observou-se um aumento na área e espessura do osso cortical e trabecular. Dados dos materiais e das propriedades estruturais óssea do grupo AC destacam os efeitos nocivos do etanol sobre as propriedades mecânicas do osso. Os resultados deste estudo demonstraram que os ratos do grupo AC apresentaram danos significativos no osso em todas as variáveis ​​analisadas. Esses resultados sugerem que a desintoxicação alcoólica é recomendada no planejamento pré-operatório e isso corrobora para o sucesso de cirurgias e reparação no tecido ósseo. ABSTRACT

The cerebral arterial circle is a polygonal shape-like arterial anastomosis placed in the brain base, whereit rounds the optic quiasm and the tuber cinereum, and also related to the interpeduncular fossa and theanterior perfurated substance. It is formed by the proximal parts of the anterior, middle and posterior cerebralarteries, and the right and left posterior communicating arteries. In order to describe the cerebral arterial circledisposition we investigated the brains of twenty rats. For each animal, the heart left ventricle was probed andacetone, distilled water at 37 °C and a solution of Neoprene Latex “450” stained with a specific red pigmentwere injected in sequence into it. To fix the brain in a better way, we isolated the head and made an apertureat the dorsal wall of the cranium and the whole specimen was fixed in a 15% formaldehyde solution. We tookoff the brain from the skull with the aid of a cold light source monocular magnifier. To take the photographicdata we used a semi-professional camera. The results showed that the cerebral arterial circle in rats is formedby branches of both internal carotid arteries and of the basilar artery, and is closed rostrally by the rostralcommunicating artery, and caudally by the right and left terminal branches of the basilar artery. We concludedfrom our data that the cerebral arterial circle in rats is compound by branches of both internal carotid arteriesand basilar artery, and that it is rostrally and caudally closed.

ABSTRACT

The pineal gland or pineal body is an endocrine gland that constitutes an important part of the neuroendocrine system, due to the secretion of melatonin, a hormone responsible for the seasonal organization of several physiologic and behavioral events of an individual’s life. Experimental researches using animals such as rats, mice and rabbits are often found in the extensive specific literature but aspects related to the morphology of mice’s pineal gland are few. Concerning its small size, the present paper performed a microscopic analysis of serial median sagittal sections of the pineal gland of 13 (thirteen) Swiss mice. The pineal gland of Swissmice was found to be in the median plane below the splenium of the corpus callosus, superior and dorsal to the habenular commissure, and rostral to the rostral colliculi. The pineal gland is closely related to the third ventricle and presents itself with a characteristic tonsillar shape with a stalk. Two types of different cells were identified in the gland, that is, astrocytes and pinealocytes, spreading randomly all over the glandular tissue. Calcifications of the pineal gland were not found in any of the observed animals.

ABSTRACT

For this study we had 10 (ten) dead bodies of dogs bearing different constitutional types. The following procedure for fixation was done in each animal: on both sides, the common carotid artery was cannulated and a 10% formaldehyde solution was then stilled. After fixation, the heads were separated from the bodies and the dissection of the target-region was performed according to classical dissection protocols, which included the removal of the skin and muscle and nerve exposition plus a further magnifying glass-aided dissection of the trigeminal nerve [V]. This study aimed to describe the anatomy of the main trunk and branching of the trigeminal nerve [V] on mongrel dogs, compare its characterization to those described in human beings and to obtain, data that conclude whether structural or branching differences of this nerve are closely related to different constitutional types of mongrel dogs.

ABSTRACT

Studies have shown that the frequency or worsening of sleep disorders tends to increase with age and that the ability to perform circadian adjustments tends to decrease in individuals who work the night shift. This condition can cause consequences such as excessive sleepiness, which are often a factor in accidents that occur at work. The present study investigated the effects of age on the daytime and nighttime sleep patterns using polysomnography (PSG) of long-haul bus drivers working fixed night or day shifts. A total of 124 drivers, free of sleep disorders and grouped according to age (<45 years, N = 85, and ≥45 years, N = 39) and PSG timing (daytime (D) PSG, N = 60; nighttime (N) PSG, N = 64) participated in the study. We observed a significant effect of bedtime (D vs N) and found that the length of daytime sleep was shorter [D: <45 years (336.10 ± 73.75 min) vs N: <45 years (398 ± 78.79 min) and D: ≥45 years (346.57 ± 43.17 min) vs N: ≥45 years (386.44 ± 52.92 min); P ≤ 0.05]. Daytime sleep was less efficient compared to nighttime sleep [D: <45 years (78.86 ± 13.30 percent) vs N: <45 years (86.45 ± 9.77 percent) and D: ≥45 years (79.89 ± 9.45 percent) and N: ≥45 years (83.13 ± 9.13 percent); P ≤ 0.05]. An effect of age was observed for rapid eye movement sleep [D: <45 years (18.05 ± 6.12 percent) vs D: ≥45 years (15.48 ± 7.11 percent) and N: <45 years (23.88 ± 6.75 percent) vs N: ≥45 years (20.77 ± 5.64 percent); P ≤ 0.05], which was greater in younger drivers. These findings are inconsistent with the notion that older night workers are more adversely affected than younger night workers by the challenge of attempting to rest during the day.

Flight safety is one of the most important and frequently discussed issues in aviation. Recent accident inquiries have raised questions as to how the work of flight crews is organized and the extent to which these conditions may have been contributing factors to accidents. Fatigue is based on physiologic limitations, which are reflected in performance deficits. The purpose of the present study was to provide an analysis of the periods of the day in which pilots working for a commercial airline presented major errors. Errors made by 515 captains and 472 copilots were analyzed using data from flight operation quality assurance systems. To analyze the times of day (shifts) during which incidents occurred, we divided the light-dark cycle (24:00) in four periods: morning, afternoon, night, and early morning. The differences of risk during the day were reported as the ratio of morning to afternoon, morning to night and morning to early morning error rates. For the purposes of this research, level 3 events alone were taken into account, since these were the most serious in which company operational limits were exceeded or when established procedures were not followed. According to airline flight schedules, 35 percent of flights take place in the morning period, 32 percent in the afternoon, 26 percent at night, and 7 percent in the early morning. Data showed that the risk of errors increased by almost 50 percent in the early morning relative to the morning period (ratio of 1:1.46). For the period of the afternoon, the ratio was 1:1.04 and for the night a ratio of 1:1.05 was found. These results showed that the period of the early morning represented a greater risk of attention problems and fatigue.

The effects of sleep disorders on the quality of life (QOL) have been documented in the literature. Excessive sleepiness and altered circadian rhythms may negatively affect ability to learn, employment, and interpersonal relations, and directly degrade QOL. The objective of the present study was to evaluate the impact of obstructive sleep apnea syndrome of varying severity on QOL. The study was conducted on 1892 patients aged 18 years or older referred by a physician to the Sleep Institute, São Paulo, with complaints related to apnea (snoring, excessive daytime sleepiness, hyperarousal, and fatigue). They were submitted to overnight polysomnography for the diagnosis of sleep disorders from August 2005 through April 2006. The patients completed the Epworth Sleepiness Scale and QOL SF-36 sleep questionnaires. They were classified as non-physically active and physically active and not-sleepy and sleepy and the results of polysomnography were analyzed on the basis of the apnea hypopnea index (AHI). The apneic subjects showed a reduction in QOL which was proportional to severity. There was a significant decrease in all domains (physical functioning, role physical problems, bodily pain, general health perceptions, vitality, social functioning, emotional problems, general mental health) for apneics with AHI >30, who generally were sleepy and did not participate in physical activities (P < 0.05). The present study provides evidence that the impact of sleep disorders on QOL in apneics is not limited to excessive daytime sleepiness and that physical activity can contribute to reducing the symptoms. Thus, exercise should be considered as an adjunct interventional strategy in the management of obstructive sleep apnea syndrome.

ABSTRACT

Numerous studies have suggested a substantial genetic contribution in the etiology of the primary form of restless legs syndrome (RLS) and periodic leg movements (PLM). We describe the symptoms, the sleep profiles and physiological parameters of two families in which several members present RLS/PLM. The proband of family 1 is a 70-year-old woman and the proband of family 2 is a 57-year-old woman; both have exhibited the symptoms since the age of 20 years. All patients in both families were diagnosed with RLS according to the criteria of the International RLS Study Group. Polysomnographic recordings were performed to quantify and to describe PLM during sleep. Sleep parameters showed decreased sleep efficiency, increased sleep latency in the arousal index and the presence of PLM in all subjects. One of the families showed an exact profile of dominant inheritance with anticipation of age at onset. In the other family, the founders were blood relatives and there was no affected member in the third generation suggesting a recessive mode of inheritance. RLS/PLM is a prevalent sleep disorder affecting about 5 to 15 percent of the population and one that substantially impairs healthy sleep patterns. Efforts to understand the underlying pathophysiology will contribute to improve the sleep and life quality of these patients.

ABSTRACT

Avaliaram-se o estado de frescor e a qualidade higiênica do pescado disponível em alguns estabelecimentos comerciais de Vila Real, uma cidade do interior de Portugal. Vinte e três amostras foram submetidas a uma análise sensorial e a análises fisico-químicas - determinação do teor de azoto básico volátil total (ABVT), do índice de refração do humor aquoso e do pH. A avaliação da qualidade higiênica do pescado foi feita por meio de diferentes análises microbiológicas na superfície e na profundidade do músculo do pescado. O pescado fresco, considerado por todos os métodos de análise, próprio para consumo, apresentou estado de frescor entre bom e satisfatório. O ABVT foi considerado o método objetivo de eleição, sempre que surgiram dúvidas relativamente ao grau de frescor do pescado. O pescado próprio para consumo apresentou valores que variaram entre os 20,60 e 27,36mgN/100g e perfil microbiológico aceitável, com contagens de microrganismos psicrotróficos entre 5,41 e 6,03 UFC/cm² na superfície e entre 4,79 e 5,28 UFC/cm² no músculo


The freshness and hygienic quality of fresh fish sold in some fish markets of Vila Real, a Portuguese town, were evaluated. Twenty-three samples were examined by sensorial and physico-chemical analyses (total volatil basic nitrogen - TVBN, aqueous humour refraction rate and pH). The determination of hygienic quality of fish was carried out by different microbiological analysis on the surface and deepness of fish muscle. Fish was considered proper for consumption by all the methods used in the evaluation of fish freshness, and was classified between good and satisfactory. TVBN was considered the objective method of election, in case of doubt about fish freshness, and fish proper for consumption presented TVBN values between 20.60 and 27.36mgN/100g. The microbiological profile was considered acceptable with psychrotrophic microorganisms counts between 5.41 and 6.03 CFU/cm² on fish surface and between 4.79 and 5.28 CFU/g in muscle deepness

ABSTRACT

The cytotoxic activity of amino (3a-e), aza-1-antraquinone (4a-e) lapachol derivatives against Ehrlich carcinoma and human K562 leukemia cells was investigated. Cell viability was determined using MTT assay, after 48 (Ehrlich) or 96 h (K562) of culture, and vincristine (for K562 leukemia) and quercetin (for Ehrlich carcinoma) were used as positive controls. The results showed dose-dependent growth-inhibiting activities and that the amino derivatives were active against the assayed cells, whereas the 4a-e derivatives were not. The allylamine derivative 3a was the most active against Ehrlich carcinoma, with IC50 = 16.94 ± 1.25 muM, and against K562 leukemia, with IC50 = 14.11 ± 1.39 muM. The analogous lawsone derivative, 5a, was also active against Ehrlich carcinoma (IC50 = 23.89 ± 2.3 muM), although the 5d and 5e derivatives showed lower activity. The interaction between 3a-d and calf thymus DNA was investigated by fluorimetric titration and the results showed a hyperchromic effect indicating binding to DNA as presented of ethidium bromide, used as positive control. The inhibitory action on DNA-topoisomerase II-a was also evaluated by a relaxation assay of supercoiled DNA plasmid, and the etoposide (200 muM) was used as positive control. Significant inhibitory activities were observed for 3a-d at 200 muM and a partial inhibitory action was observed for lapachol and methoxylapachol.

Efeitos de estádio da lactação, idade à classificação e grupo contemporâneo de rebanho, ano e estação de classificação foram considerados nas análises de 21 características lineares de tipo e pontuação final de 2.122 animais da raça Holandesa, filhos de 178 reprodutores e nascidos em 83 rebanhos. Utilizou-se o método dos quadrados mínimos nas análises de variância das características. O efeito de estádio da lactação teve influência sobre as características do sistema mamário, evidenciando que houve alterações morfológicas durante o período de lactação para todas as características de úbere, exceto inserção do úbere anterior, textura do úbere e comprimento dos tetos. Dentre as características de conformação/capacidade corporal, profundidade corporal foi a mais influenciada pelo estádio da lactação, seguida do tamanho corporal, largura torácica e força lombar. Para todas as características de conformação/capacidade corporal o efeito da idade à classificação revelou-se altamente significativo. Dentre as características lineares da categoria pernas/pés, a variável idade à classificação foi importante (P<0,05) apenas para vista lateral das pernas. O efeito de idade sobre as características de garupa variou consideravelmente, sendo importante apenas para largura (P<0,01). Pontuação final e angulosidade também foram influenciadas (P<0,01) pelo efeito da idade. A verificação de significância estatística indicou a existência de componentes de meio e evidenciou a importância do estudo desses componentes.

ABSTRACT

Correlações genéticas e fenotípicas entre 21 características lineares de tipo e produção de leite em até 305 dias de lactação foram estimadas pela máxima verossimilhança restrita sob modelo animal, a partir de dados da Associação de Criadores de Gado Holandês de Minas Gerais. Nas análises das características lineares, os efeitos fixos utilizados no modelo incluíram estádio da lactação, idade do animal, grupo contemporâneo de rebanho, ano e estação de classificação, e o efeito aleatório do pai da vaca. As características lineares que apresentaram maiores correlações genéticas com produção de leite foram largura do úbere posterior (0,60) e largura de garupa (0,37), indicando não haver antagonismo genético entre a seleção para essas características e a produção de leite. As correlações fenotípicas entre características lineares de tipo e produção de leite foram, em geral, menores do que as correlações genéticas correspondentes. As correlações fenotípicas entre as características lineares apresentaram-se altas dentro das seções de conformação/capacidade e de sistema mamário. Correlações genéticas entre algumas características lineares foram altas, possibilitando a exclusão de algumas delas do programa de classificação linear adotado para a raça Holandesa no Brasil.

ABSTRACT

Propõe-se um método de monitorização hemodinâmica que compreende a avaliação do debito cardíaco, pressões atriais, pressão arterial, frequência cardíaca, temperatura corporal e volume urinário. Esta monitorização mais ampla foi utilizada em 12 crianças com media de idade 24 meses (desvio padrão 17 meses). As principais indicações foram: idade e peso reduzidos, necessidade de drogas inotrópicas ao sair de circulação extracorpórea, complexidade da cardiopatia e da correção cirúrgica, e insuficiência cardíaca ou respiratória antes da operação. São analisados os resultados terapêuticos, através de curva de função ventricular e particularidades na evolução hemodinâmica no pós-operatório em gráficos separados. O debito cardíaco foi obtido por termodiluição e são discutidos aspectos relacionados com o método utilizado em sua determinação. O procedimento revelou-se seguro e reprodutível, fornecendo subsídios importantes para decisões terapêuticas

ABSTRACT

O prognatismo mandibular e uma das deformidades faciais mais frequentes e acarreta muitas vezes graves problemas de ordem funcional e psicologica para os seus portadores. Embora varias tecnicas cirurgicas venham sendo propostas desde o seculo passado para a correcao desta deformidade, a literatura mundial denuncia a ausencia de uma tecnica que satisfaca plenamente cirurgiao e paciente. Isto se deve ao grau de morbidade do tratamento que acarreta algumas vezes cicatrizes na face, neuropatias de resolucao demorada, aparecimento de sindrome articular ou variadas formas de ma-oclusao incluindo a recidiva da deformidade. Uma revisao tardia de 50 casos de prognatismo nos quais a osteotomia sagital bilateral do ramo por via intra-oral a Trauner e Obwegeser foi realizada revela os sucessos e insucessos que obtivemos com este procedimento cirurgico que a nosso ver e o melhor. Aspectos pre, trans e posoperatorios sao discutidos, bem como os fatores determinantes de nossas complicacoes e sequelas
